  Amr Amin completed his PhD at the University of Illinois at Chicago and received a post doctoral training in the field of molecular genetics at the University of Pennsylvania School of Medicine. He started his academic career at UAE University where he serves now as a Full Professor of Cell and Molecular Biology. His research focuses on ways to control cancer, particularly liver cancer. He published many research articles and reviews and serves as a regular reviewer and as an editorial member of many specialized peer reviewed journals.   Gene and Stem Cell Therapy For Lung Injury: In Vivo Reprogramming of Alveolar Epithelial Cells

  Abstract

  Chronic obstructive pulmonary disease (COPD), is a major public health problem but currently without any effective pharmacologic approach for the treatment. Stem cell therapy based on transplantation of in vitro propagated stem/progenitor cells may be a feasible approach for COPD treatment. However, due to the complexity of cell source and lung microenvironment, whether transplanted cells have differentiated for reconstitution of airway/alveolar epithelium were questioned. Furthermore, safety issues have raised concerning the use of stem cells in vivo. Our lab has focused on in vivo gene delivery of stemness genes in somatic lung epithelial cells using PEI nanoparticles for lung injury treatment. In mouse model of elastase-induced emphysema or bleomycin-induced fibrosis, we found that the transient gene delivery of a gene (indicated as COP-X1 here) in alveolar epithelial cells post-injury induced efficient regeneration of alveolar epithelium and improved pulmonary function. The regenerated regions showed normal alveolar epithelial phenotype and extracellular matrix components, without the symptoms of neoplasia. Ex vivo culture of COP-X1 targeted alveolar epithelial cells formed alveolar-like spheroids more efficiently than control cells. NGS analysis suggested the potential in vivo reprogramming of transfected cells. In summary, our study suggests that in vivo reprogramming of somatic cells mediated by stemness gene delivery could be a feasible approach for tissue regeneration. In vivo gene delivery holds promise for the future treatment of lung diseases such as ARDS or COPD in clinic.

  Professor Wu, currently, is Academician of Academia Sinica, and Distinguished Chair Professor of National Yang-Ming University in Taiwan. He was a pioneer in biophysical approach to molecular biology, best known for his work on presenting biophysical evidence of the 4-step mechanism of gene transcription, and the discovery the presence of zinc ions in eukaryotic transcription factor TFIIIA, the first example for the class of proteins known as zinc finger proteins. After returning to Taiwan Professor Wu made many important contributions to the studies of cancer metastasis in lung cancer, and of acute and chronic Lung injuries such as ARDS and COPD, two of the serious health threats to the Taiwanese population, with important clinical significances. Recently, he is interested in applying stemness concept for analyzing tumor progression in lung cancer as well as gene and stem cell therapies of lung injuries.   The use of Bioquantine and allogeneic Mesenchymal Stem Cells combined with a spinal cord stimulation system in a No Option patient with ASIA-A scale

  Abstract

  In 1973 the American Spinal Injury Association made the International Standards for Neurological Classification of Spinal Cord Injury (ISNCSCI). In this clinical review our patient was classified after the vertebral fixation surgery with a ASIA-A scale injury after suffering a fracture and luxation at T-12-L1, having total spinal cord section (Fig.A). Based on the research made by Sergei Paylian PhD on animal models [1] and the safety use of allogeneic MSCs demonstrated on multiple animal models applications [11,12], we decided to apply a experimental translational medical protocol based the research and the previous outcomes obtained by Hamid and MacEwan [8,9] and decided to customize it exclusively to our patient based on the clinical evidence and personalizing the therapy on evidence. The medical team designed an ambulatory method utilizing a C-arm to apply the allogeneic MSCs in situ and using a intrathecal (subdural) catheter using a slow pump release system for the rest of the biological material with an optimum tolerance and minor side effects (mild fever, miyalgias and headache) on the first 48hrs hour after application. The experimental use of mRNA Bioquantine® was well tolerated with its purified form (intra and extra-oocyte liquid phases of electroporated oocytes [6]) showing to be well tolerated by the patient without any anaphylactic reaction. The current clinical report is meant to demonstrate the beneficial changes with the use of Bioquantine® and its administration in a patient with a severe SCI offering a possible optional therapy and potential neuroregeneration in this clinical condition. Mesenchymal stem cells (MSCs) are ideal for cell-based therapy in various inflammatory diseases because of their immunosuppressive and tissue repair properties. Moreover, their immunosuppressive properties and low immunogenicity contribute to a reduced or weakened immune response elicited by the implantation of allogeneic MSCs compared with other cell types, allogeneic MSCs are a promising option because of their low immunogenicity and immunosuppressive and tissue repair capabilities. The positive findings throughout the evolution of our protocol for spinal cord injury with the obtained results at this stage is a promising scientific based and evidence based medicine protocol that can be offered in the near future as an option for severe SCI patients. The functionality of the RestoreSensor® SureScan® by providing the electric stimulation fortifies the medical outcome and has given the patient the confidence to perform his physical rehabilitation with more energy for a longer time by the increase or decrease of the intensity of it according to the type of exercise regulated by the control-battery he handles. At this date, after 8 intrathecal applications of allogeneic MSCs and Bioquantine® in situ combined together we have got the following outcomes: an improvement in sensitivity, strength in striated muscle and smooth muscle connection by increased muscle mass (Fig.B) and sphincter control, at 23 months after the first regenerative therapy and 12 months after the placement of RestoreSensor® the patient is showing an evident improvement on his therapy of physical rehabilitation (legs movement and control of them) having the following movements reported by the physical therapist: a)hip: adduction and external rotation, extension, abduction, internal rotation; b)knee: flexion; c)toe: MP and IP extension, also reporting an easier and functional crawling forward and backwards, and since 3 months ago the patient is capable to stand on his knees for 2 or more minutes without any support (Fig. C) and taking small steps on his knees forward and backwards for the first time in his process, showing a progressively important functionality on both limbs, voluntary movement at both feet and an increase in sensory perception.

  | CRISPR/Cas9-mediated mutagenesis of CCD8 gene in tomato (Solanum lycopersicum Mill.) provides resistance to parasitic weeds

  Abstract

  Broomrapes (Phelipanche and Orobanche spp.) are obligate holoparasites that attack the roots of many economically important crops throughout the semiarid regions of the world, especially the Mediterranean and Middle East. The parasite seeds have strict requirements for germination, involving preconditioning and exposure to specific chemicals (strigolactones (SLs) exuded by the host roots. SLs are plant hormones derived from carotenoids via a pathway involving the carotenoid cleavage dioxygenase genes CCD7 and CCD8. Having no effective means to control parasitic weeds in most crops, here we demonstrate that CRISPR/Cas9-mediated genome editing of the CCD8 gene can be used to develop host resistance to the parasitic weed P. aegyptiaca. Cas9/single guide RNA construct was targeted to the second exon of CCD8 in tomato (Solanum lycopersicum ) plants via Agrobacterium transformation. Several mutated tomato lines with variable deletions in CCD8 were obtained with no identified off-targets. Although, mutant lines had morphological changes (excessive shoot branching and adventitious root formation) but some lines showed a significant reduction in parasite infestation compared to non-mutated tomato plants. Considering the impact of parasitic weeds on agriculture, the current study offers insights into the development of a new efficient and environmentally friendly method for parasitic weed control.

  | The effectiveness of systemic treatment of breast cancer depending on the body weight index using levocarnitine.

  Abstract

  The prevalence of breast cancer (BMD) in the world in general and in Ukraine is steadily increasing. Epidemiological, experimental and clinical studies have shown that metabolic disturbances associated with body mass index (BMI)> 30 kg / m2 increase the risk of occurrence and worsen the clinical course of breast cancer. Thus, in patients with obesity, a decrease in the sensitivity of the tumor to systemic antitumor therapy, an increase in the frequency of postoperative complications and a decrease in the rates of general and non-recurrent survival. The aim of the study was to improve the results of neoadjuvant systemic antitumor therapy in breast cancer patients with abdominal obesity (BMI greater than 30 kg / m2) by administering levocarnitine in combination with NSAT for the correction of metabolic disorders as the main pathogenetic part of obesity. For the study used a retrospective study between 2010 and 2014 three hundred patients (prevalence of 12.4% which is 100 thousand. population in the Dnipropetrovsk region) with BMI> 30 kg / m2, morphologically verified diagnosis of different forms of breast cancer and all stages (I- IV). Subsequently, a group of comparisons with abdominal obesity BMI> 30kg / m2 with a definite molecular subtype of tumor, levels of expression of estrogen receptor ER, progesterone PgR, Her-2 / neu, Ki-67 proliferation index was formed. The observation group of patients with breast cancer and BMI> 30kg / m2 was formed in the period from 2014 to 2018 due to prospective observation of "case-control". Thus, the study involved 108 patients aged 32 to 76 years (mean age (58 ± 2). With nodal breast cancer II-III stage. As a result of randomization of all patients (n = 108) on breast cancer with BMI> 30 kg / m2, depending on the appointment of levocarnitine during NSAT, were divided into 2 groups: comparison and observation. In the comparison group, patients (n = 58) with BMI> 30 kg / m2 patients with breast cancer who did not receive levocarnitine during NSPT, and in the, observation group - patients (n = 50) on breast cancer with BMI> 30 kg / m2 who received levocarnitine during NIST. For the first time, on the basis of a comprehensive analysis of the results, it was estab-lished that the appointment of levocarnitine in patients with breast cancer with BMI> 30 kg / m2 contributes to increasing the clinical and morphological efficacy of neoadjuvant systemic antitumor therapy (NSAT). It was found that in patients with breast cancer with BMI> 30 kg / m2, who were prescribed levocarnitine, organosurgery surgical interventions were significantly more frequent. The algorithm of diagnosis and treatment of breast cancer in patients with obesity has been developed and scientifically substantiated. A theoretical generalization of modern scientific data on the influence of metabolic disorders that are characteristic of obesity, on carcinogenesis of breast cancer is carried out. A complex (integrated) scheme of mechanisms of influence of molecular disorders associated with obesity, on carcinogenesis of breast cancer is developed. It is determined that the leading role in carcinogenesis is played by: abdominal obesity, hyperglycemia, dyslipidemia and chronic subclinical inflammation. The carcinogenic effect of these factors is realized due to the ability of the adipose tissue of the mammary glands to increase the local concentration of estrogen by peripheral aromatization of androgens, the mitogenic effect of metabolic disorders on the epithelium of the mammary gland. It was established that in patients with breast cancer with BMI greater than 30 kg / m2 receiving levocarnitine, a statistically significant increase in the number of cases of clinically complete regression of tumors was observed at 12.8% (5.2% vs. 18.0%; p <0.05) and a partial regression of 13.1% (6.9% vs. 20.0%, p <0.05). Lowering the frequency of breast cancer progressing on the background neoadjuvant systemic anticancer therapy in patients with a group of observations by 44.3% compared with women with the comparison group (16.0 vs. 60.3%, p <0.05). According to the results of analysis of the parameters of cellular, humoral immunity (CD3, CD4, CD8, IgG, IgM, IgA) and proinflammatory cytokines (IL-6, TNFα), there was a decrease in the number of patients with breast cancer with BMI greater than 30 kg / m2 in (NSAT) Levocarnitine with regression rates was used for juvenile systemic anticancer therapy, namely: for CD3, which was 41.4%; for CD4, which was 50.0%; for CD8, which was 23.5%; for IgG, which was 40.4%; for IgM, which equaled 48.7%; for IgA, which was 44.4%; for IL-6, which was 51.1%; for CD3, which was 50.0%, respectively. It was found that the appointment of levocarnitine in patients with breast cancer with BMI greater than 30 kg / m2 during preoperative systemic antitumor therapy increases its efficacy compared with patients who received neoadjuvant systemic antitumor therapy. Registered increase in the incidence of clinically significant responses (CR + PR) for the treatment in 63.2% of patients in the observation group who were administered in the treatment Levocarnitine (28.0% vs. 12.1%, p <0.05) compared with a comparison group. Based on the research, it was proved that in the group of patients with BMI greater than 30 kg / m2 receiving levocarnitine, there was a statistically significant increase in the frequency of performing functionally justified organoleptic operations by 53.8% compared with the comparison group (27.1% vs. 10.9%; p <0.05). Also, the administration of levocarnitine during neoadjuvant systemic antitumor therapy increases the number of cases of complete morphological regression (V degree of curative pathomorphosis) by 77.8% in the observation group compared to patients from the comparison group (18.8% vs. 3.6%; p <0.05). Found that neoadjuvant systemic anticancer therapy in breast cancer patients with a BMI over 30 kg / m2 does not affect the degree of differentiation and histological type of tumor and the expression of Her2 / neu (p> 0.05). After neoadjuvant systemic anticancer therapy, regardless of the purpose levocarnitine in decreasing residual tumor cell proliferation index (Ki-67) and increased incidence of Luminal A molecular type of breast cancer. This may indicate that neoadjuvant systemic antitumor therapy leads to the death of the most aggressive tumor clones. It was investigated that the appointment of levocarnitine in patients with breast cancer with BMI greater than 30 kg / m2 does not affect the indicators of general and non-recurrent survival, with observation (18,1 ± 1,5) months. These data are in the basis for integrated assessment and in-depth understanding of the negative impact of abdominal obesity associated with impaired carbohydrate and fat metabolism in carcinogenesis and criteria can be aggressive course of breast cancer and adverse prognostic factors. Correction of metabolic disorders caused by obesity, eliminates their negative impact on breast cancer carcinogenesis and prognosis of the disease, allowing you to customize and personalize the treatment strategy for breast cancer patients with a BMI> 30 kg / m2 and improve its results. When planning treatment in patients with breast cancer advisable to consider the presence of symptoms and diseases that are unique and are associated with obesity where BMI over 30 kg / m2, as the presence of disorders of carbohydrate and fat metabolism and hypertension is an indication for additional surveys aimed at identifying metabolic disorders with the aim of individualizing and personalizing tactics of systemic antitumor therapy. Against neoadjuvant systemic anticancer therapy for breast cancer patients with a BMI over 30 kg / m2 to be additionally administered drugs that increases the incidence of complete morphological regression. This drug is levocarnitine in therapeutic doses (1500 mg / day), which has proven to be an effective means to increase the number of cases of clinically relevant responses (CR + PR) to the treatment. Analysis and systematization of the results of the study allowed to create algorithms of diagnosis and treatment from the standpoint of personalization and individualization, based on which there is a correlation dependence that can be used in the clinical practice of oncologic hospitals to improve the treatment of patients with breast cancer with BMI greater than 30 kg / m2. Thus, the use of neoadjuvant systemic antitumor therapy in combination with levocarnitine contributes to an increase in the number of cases of objective clinical (complete regression and partial regression) and morphological (therapeutic pathomorphosis IV and V degree) tumor response to cytotoxic breast cancer therapy in patients with BMI> 30 kg / m2 and frequency of organ-saving surgical interventions.   | In vitro study of porphyrin photosensitizers for cancer therapy

  Abstract

  Photodynamic therapy (PDT) is based on activation of photosensitizers (PS) by light delivery to generate reactive oxygen species, basically singlet oxygen (SO), damaging cell membrane. We studied in vitro the processes involved in photodynamic therapy on a model bilayer lipid membranes (BLM) by measuring the boundary potential applying the Intramembrane Field Compensation (IFC) Method (Sokolov and Kuz’min, Biofizika, 25:170, 1980). IFC method allowed to monitor binding of PS on BLM and damage of di-4-ANEPPS, which was used as target molecules (TM) of SO under illumination. The rate of oxidation of TM was calculated from the kinetics of the potential drop under illumination and its restoration in dark. We studied the adsorption and photodynamic efficiency of newly synthesized positively charged porphyrins, namely -imidazolyl substituted porphyrin and it’s Zn(II) and In(III) complexes; and two phosphorus (V) complexes of meso-(p-pyridyl)-triphenylporphyrin bearing hydroxyl and ethoxyl axial ligands. We observed the adsorption of these PS on BLM by measuring the boundary potential change, which was proportional to the logarithm of concentrations of each compound. To evaluate their photodynamic efficiency, we determined the rate of oxidation (R) of TM adsorbed either in the same or opposite surface of the BLM where molecules of PS were adsorbed. Similar R values were obtained suggesting equal distribution of SO between two sides of BLM. In conclusion, the adsorption of PS compounds on BLM was found to be a main factor influencing on the photodynamic efficiency of the porphyrin-based PS used in this study.

  Abstract

  The tea plant,Cameliasinensisis native in South East Asia, and is now cultivated in more than 30 countries. Green tea is produced by steaming freshly harvested tea leaves. This causes the inactivation of fermentation enzymes,so that the green color of the leavesis maintained. Green tea has been associated with the prevention and reduction of a wide range of severe health conditions such as cancer,cardiovascular diseases, malariaand Alzheimer disease. The health benefits associated with green tea consumption has been predominantly attributed to green tea polyphenol-EGCG, (-)-epigallocatechin gallate. The cytotoxicity of green tea was explained by its anti- oxidant activity and byinterfering with signal transduction processes.In the past, leaves of Camelia sinensis were inserted directly into boiling water and the subsequent tea was consumed. In the present time, extracts are prepared from the leaves, added to water and consumed.After identifying the active compounds as polyphenols, they were swallowed directly as medicinal powders. Unfortunately, factors like temperature, pH and oxygen, reduced the activity of the powder,and the occurrence of unpleasant taste and the degradation by intestinal tract enzymes, hampered their use. In the future,nanoparticles will be used as delivery vehicles of ECGC for cancer therapy. These stable encapsulatedpolyphenols can be added to food. It may provide a new therapeutic technologyto the use of green tea for the benefit of man.

Herscientific work includes research in ethno botany of the Middle East, and the effect of the environment on the quality of natural compounds.She received her PhD from ColumbiaUniversity in NY, USA and worked as a senior scientist at the AgricultureResearch Center of Israel. For 10 years (1991-2001),she served as the head of The Department of International Collaboration of the Agriculture research Center in Israel. Shepublished many scientific papers and books. Shewas a Visiting Professor at various universities in the USA, Europe and China. She is one of the founders, and a member of the board of the Israeli Society of Medicinal Plants.In addition, she is teaching courses on Medicinal Plants in the Universities and to the public.
